What happened, in summary

A 58-year-old woman with lung adenocarcinoma had already received several systemic treatments before entering the TAPUR precision-medicine study. Her prior therapy included carboplatin and pemetrexed chemotherapy, durvalumab, and later nivolumab, with progression on nivolumab.

Genomic testing identified a CD74-NRG1 fusion in her tumor. Based on that finding, she started the targeted drug afatinib in December 2020. Her measurable tumors decreased by about 31%, meeting criteria for a partial response. The response lasted 24 weeks, and the cancer progressed after 32 weeks on afatinib.

No major treatment-related adverse events were recorded during the study treatment. She died in April 2022. Her case was one of several rare NRG1-fusion cancers treated with afatinib in the TAPUR study and demonstrated that a genomic finding can sometimes identify a treatment option even after multiple previous therapies.
